The sizes of the Î» / EcoRI DNA fragments, in base pairs, are 21226, 7421, 5804, 5643, 4878, and 3530. A sample volume of 20 Î¼l is recommended for ethidium bromide or SYBR Safe staining and 35-40 Î¼l is optimal for methylene blue staining. Shortly before loading, heat the fragments at 65°C for 3 minutes and quickly chill on ice. Expt. 709
